Recently, I hosted a webinar which discussed what surface zeta potential is and how it can be measured. The webinar attracted huge interest and it was a pleasure to see so many people attending and asking some excellent questions. In fact, there were so many questions asked, that we ran out of time at the end of the webinar and had to answer many of them via e-mail. Some of these are listed below.
The webinar discussed the zeta potential of solid, planar surfaces (rather than colloidal dispersions or molecular solutions) …

How can I use DLS for sample xyz?
What sample concentration / refractive index is needed for reliable DLS size measurements?

Has it been done before?
In Dynamic Light Scattering (DLS) intensity fluctuations in the sample are analyzed to obtain size information on the sample. The technique is applicable to a wide size range from sub-nanometer up to a few microns. In order to obtain a meaningful result, a minimum scattering signal strength must be present. Malvern’s Zetasizer Nano software contains a calculator to help estimate the required minimum concentration.
If anyone else has …

The NEW MicroCal PEAQ-DSC – the gold standard for thermal stability determination
Differential scanning calorimetry (DSC) is an established biophysical tool for protein stability characterization during drug development. DSC data are used to advance the most stable and ‘developable’ proteins into the pipeline, and optimize the process and formulation conditions which will maintain protein stability during manufacturing and storage. DSC data are also important in higher order structure (HOS) protein characterization, lot-to-lot comparability, and characterization of biosimilar drugs compared to innovators.
Our previous technology, the MicroCal VP-Capillary DSC Automated system, is a …

Recently, I’ve presented a new webinar focusing on the protein characterization using the OMNISEC, the highest quality Gel Permeation Chromatography (GPC) / Size Exclusion Chromatography (SEC) system for the measurement of absolute molecular weight, molecular size, intrinsic viscosity, branching and other parameters. Thank you to all participants for your interest and attention during the webinar.
In this webinar, I highlighted the key features and benefits of the OMNISEC system. How to run samples and analyze data and how to access repeatability of multiple sample measurements.

As usual, after the live section of …

DSC for the regulated environment

Advancing biotherapeutic molecules along the drug development pipeline is a complex, time-consuming and expensive process. Good stability is key to a candidate’s progression, and must be assessed and reviewed throughout development – from candidate selection, through formulation development, all the way to batch-to-batch comparability or biosimilarity assessment….and beyond! Objective, reliable stability data is needed for confident decision-making about which drug products and processes to move through development into commercialization.
Differential Scanning Calorimetry (DSC) is a biophysical tool for protein stability characterization. The MicroCal VP-Capillary DSC system, launched in 2002, …

The Pharmaceutical Analysis of Powders – Particle Size Determination seminar is to be held March 1st at Bella Sky in Copenhagen and is co-organized by Malvern Instruments and Particle Analytical.
If you are working with products manufactured from powdered solids in the pharmaceutical industry, this might be the course for you.
The most important physical property of particulate samples is particle size – this has a direct influence on material properties such as reactivity, dissolution rate, stability, flowability and handling. Therefore, particle size distribution should be known, and also controlled. Ultimately this helps to …

Why measure a viscosity flow curve, rather than just give a number?

Very often chemists are asked to provide a single viscosity value for a product or formulation without much information about the required conditions of the measurement.  For an oil or low viscosity liquid which is Newtonian this is pretty straightforward, so long as your viscometer is properly calibrated and the measurement temperature is provided. However, most industrially interesting or complex fluids such as polymer solutions, suspensions and emulsions are non-Newtonian, meaning that their viscosity varies with shear rate – …

Electrophoretic mobility measurements from proteins: How to protect proteins in an electric field

What is the protein mobility mode?
When applying an electric field to a sample containing protein molecules, there are a few considerations to take into account to avoid “over-stressing” delicate biomolecules. The force a charged object experiences in an electric field is proportional to its charge times the electric field. Therefore the electric field strength (defined by the applied voltage divided by the distance between the electrodes, in V/cm) is more relevant than the absolute value of the applied …

How to measure it with DLS?
What virus concentration is needed for reliable DLS size measurements?

What is a virus?
In cell biology, a virus is a self assembly of organic molecules (mainly proteins) in the size range 20 nm – 300 nm (diameter). Viruses hold great interest for human gene therapy researchers, and the AAV-2  virus shown to the left is an example.
How to size a virus in solution?

One of the techniques available to size viruses (as well as other biological nanoparticles) is Dynamic Light Scattering (DLS), where intensity fluctuations in a small …

Paul Kippax alerted me to an interesting piece in Chemical Watch (Global Risk and Regulation News). It’s based on a comment by the European Commission Joint Research Council (JRC) that:
‘The term “particle size” does not sufficiently describe the exact quantity that is measured, across different nanomaterial particle size analysis (PSA) methods’.
‘Our findings show that the equivalent diameter values of some measurement methods do not agree and that the results of some others only agree, due to their relatively large uncertainties,” the study authors say.
“For a reliable comparison of particle size, …

Peeling back the foil on a gold coin. Popping a perforated window on a festive calendar. Pursuing your favourite coloured wrapper around a bowl. The prize inside many little packages we open this season will be chocolate.
The secret of chocolate’s almost universal appeal is in the mouthfeel the “snap” of the first bite, how the chocolate melts and how it coats the inside of the mouth. Despite being a consumer experience, mouthfeel is strongly related to materials science – chocolate being a complex composite material.
Our instruments hold the key to many of …
