Archimedes is a new, highly innovative instrument which uses the technique of resonant mass measurement to detect and accurately count particles in the size range 50nm – 5µm, and reliably measure their buoyant mass, dry mass and size. Particularly useful for the characterization of protein aggregates in formulation or buffer.

I know this sounds like the beginning of a bad joke, but in this case, it was actually the beginning of the 2017 Colorado Protein Stability Conference, held in July at Beaver Run Resort in Breckenridge, Colorado. Last week my kids got very frustrated with warm French toast and a new brand of chocolate spread. The French toast, was …of course…warm, and the chocolate was misbehaving: melting and dripping on their fingers …

Recently this question was posed where a current dynamic light scattering (DLS) user inquired about detecting large but subvisible particles in a formulation:
“I have been looking at solutions from prefilled syringes which appear in flow microscopy to have significant levels of oligo-micron-sized silicone oil droplets. I don’t see any indication of these in my DLS measurements.”
With specifications for DLS systems like the Zetasizer from sub nanometer to several microns the initial concern is not unreasonable. However, on closer look the situation is not so apparent.  The silicone oil droplets …

PhD student Polly came into the lab on Monday morning to find that her purified protein had started to aggregate after a weekend in the fridge. The profile she got from her Dynamic Light Scattering (DLS) measurement clearly showed two peaks, with the second in the micron range. Her supervisor suggested she tried Nanoparticle Tracking Analysis (NTA) to get more information on the different sized aggregate species in her sample. This showed that there were aggregates of many different sizes present. The question was, what should she do next?

Last week I attended the 4th Microbubble Symposium hosted by the Leeds Microbubble Consortium at University of Leeds in the UK. The meeting focused on the Fabrication, Characterization and Translational Applications of Microbubbles. At the end of this month, I am off to London’s Hammersmith for a couple of days to attend Oxford Global’s 6th Annual Proteins Congress.  This meeting wil be held on March 25th-26th at the Novotel London West Hotel, and promises to attract upwards of 220 scientists working within the proteins and biotherapeutics worlds.  The focus of the meeting, which is co-located with the Biosimilars & Biobetters Congress and the 3rd Annual Vaccines Congress, will be on antibodies, biotherapeutics, protein expression, purification and characterization, and immunogenicity and aggregation.
